DT2 PROBLEM - PLEASE HELP

Hello, I have recently with a friend completely rebuilt a 1972 yamaha dt-2. But the darn thing won't start. It ran once but woiuld not shift up so we fixed the shifting problem and it hasn't run since 'we just adjusted a screw' It has good spark and we've rebuilt 2 carbs and tried everything, done the timing, we have exhausted all knowledge. Does anyone out there know if there is some trick to getting these things to start.

Any help would be much appreciated.
We are at our wits end.

Start with the basics here. Do you have a new spark plug? Is there fuel on the plug when you kick it over for a while? Is there and audible snap when you kick it over with the spark plug against the cylinder? If it ran before you "adjusted the screw" then it seems to me that you did something while you were fixing that. Check your grounds while you are there.
